The purpose of this project is to assess loss of muscle mass in different mouse models of dysferlinopathy. The tibialis anterior, triceps surae complex, psoas and gluteal complex will be dissected bilaterally from the following mouse strains – C57Bl/6J, BlaJ, A/J, A/J Dysf+, SJL/J and KI32. We expect that dysferlin deficient muscles will exhibit a time dependent decline in muscle mass and total protein, with an effect order of psoas > gluteal complex > gastrocnemius complex > tibialis anterior. We expect that dysferlin competent muscles will show no reduction in muscle mass. We also expect that TSP-1 levels will negatively correlate with muscle mass, such that TSP-1 levels will be highest in the most severely affected muscles.
