Recent studies have demonstrated that dysferlin is expressed in primary granules and secretory vesicles of neutrophils. We have conceived a simplified diagnostic assay that involves the use of immunohistochemistry on blood films to detect dysferlin expression in these cells (https://doi.org/10.1016/j.nmd.2019.09.008). This assay uses as little as one drop of blood. Neutrophils constitute 50-70% of the leucocyte population and dysferlin expression can be assessed on a peripheral blood film (PBF) by immunohistochemical (IHC) techniques. This study validated the assay for diagnosis through a blind study on a large Indian cohort of genetically diagnosed dysferlinopathy patients and healthy individuals.
